Gerhard Wagner studierte Physik an der Technischen Universität München und schloß das Studium 1971 mit dem Diplom ab. Die Diplomarbeit in Biophysik unter der Betreuung von Adalbert Mayer und Fritz Parak befaßte sich mit Mößbauer-effekt-Studien an den eisenhaltigen Proteinen Hämoglobin, Myoglobin und Ferredoxin. Von 1972 bis 1977 führte Gerhard Wagner eine Doktorarbeit an der Eidgenössischen Technischen Hochschule (ETH) in Zürich aus. Unter der Leitung von Kurt Wüthrich untersuchte er mit Kernresonanzmethoden die molekulare Dynamik von Proteinen. Es wurde unter anderem gezeigt, daß aromatische Seitenketten selbst im Innern von Proteinen schnell rotieren. Nach einem kurzen Aufenthalt am Massachusetts Institute of Technology (MIT) im Labor von John Waugh (1978–1979) kehrte Gerhard Wagner an die ETH Zürich zurück. Er entwickelte NMR-Methoden zur Resonanzzuordnung und Strukturbestimmung von Proteinen. Dies beinhaltete eine erste fast vollständige Resonanzzuordnung eines Proteins (BPTI). 1987 nahm er eine Berufung als Associate Professor an die University of Michigan (Ann Arbor, Michigan, USA) an. Hier entwickelte er Triple-Resonanzmethoden zur Resonanz-zuordnung von isotopenmarkierten Proteinen. 1990 wurde er an die Harvard Medical School (Boston, Mass., USA) berufen. Er ist gegenwärtig der Elkan Blout Professor for Biological Chemistry and Molecular Pharmacology. Seine Forschungsinteressen sind Struktur-Funktionsbeziehungen von Proteinen bezüglich Aktivierung von T-Zellen, Translationsinitiation in Eukaryoten und Apoptosis. Zusätzlich befaßt er sich mit Metabolomics-Methoden zum Studium von menschlichen Krankheiten.
